In this invention, a novel and efficient method was developed and can easily and quickly unveil the total microbial diversity information from soil and environment samples. Use of 30?g?300 MPa super high pressure can destruct large amount of sample in one time and release the whole DNA. The whole environmental DNA were then PCR amplified by bacterial specific primers for 16s rDNA V3 region and fungal specific primers for 18s rDNA NS2 region, respectively. Through denaturing gradient gel electrophoresis (DGGE) separation and study??up to 66%??50% of bacterial diversity rich and up to 166%??133% of fungal diversity rich can easily obtained. The results clearly demonstrate the superiority of this novel innovative method in the study of environment microbial diversity.
